class PreCultureSource(*values)[source]

What the screened culture was inoculated FROM.

  • frozen_stock: straight from a frozen pool, no pre-culture (Hillenmeyer’s negative generation counts: “taken directly from the freezer”).

  • overnight_culture: an overnight culture of unstated phase.

  • log_phase_culture: grown to log phase before treatment (Hillenmeyer’s positive counts: “grown overnight until log phase (OD600= 2.0)”).

  • thaw_recovery: thawed and recovered briefly in medium (Hoepfner HOP: “thawed and recovered for 3 h in YPD”).

  • timed_culture: grown for a stated time (PreCulture.duration_hours) whose growth phase the source does not state (De Chiara 2022: a 24 h YPD culture before transfer into PBS, a 48 h presporulation culture before sporulation medium). Distinct from overnight_culture because the duration is stated and is not overnight.
